May and colleagues combine proximity labeling with quantitative mass spectrometry to profile the proteome of primary cilia in cells responding to Hedgehog. They identify signaling factors that undergo dynamic ciliary enrichment and uncover an unanticipated regulation of ciliary cAMP signaling.
Hellyeh Hamidi and Johanna Ivaska (University of Turku) preview work from the Korolchuk lab (https://bit.ly/3aUpDy2) describing focal adhesions as hubs mediating growth factor signaling via mTORC1 and amino acid input into the cell.
Alexandre T. Vessoni, Luis F.Z. Batista (Washington University in St. Louis) and colleagues demonstrate telomere shortening leads to a unique DDR response in human pluripotent stem cells. Unlike terminally differentiated cells, telomere shortening induces formation of single-stranded DNA telomere overhangs in hPSCs, which activate ATR signaling and lead to mitotic catastrophe and p53-dependent cell death.
Harinath Doodhi, Tomoyuki U. Tanaka (University of Dundee) and colleagues reconstitute yeast kinetochore-microtubule interface in vitro and compare the relative strength of distinct kinetochore-microtubule interaction modes. Their results suggest how Aurora B kinase promotes the exchange of kinetochore-microtubule interactions to eliminate aberrant interactions and establish chromosome bi-orientation.
Yumiko Oshima, Mariusz Karbowski (University of Maryland School of Medicine Official Page) and colleagues report that in the absence of #Parkin activity cells adapt to mitochondrial proteotoxicity through a Drp1-mediated mechanism that involves severing of the OMM and IMM ubiquitination, followed by recruitment of autophagy machinery to the ubiquitinated IMM proteins.
In our latest Viewpoint, Christopher Burd (Yale School of Medicine) discusses recent studies that implicate the GRASP65 and GRASP55 proteins in organizing stacks of Golgi cisternae into a ribbon.
